Immunogen

This STAT5A/B phospho Tyr694/Tyr699 antibody was raised against a peptide containing phospho Tyr694 of human STAT5A. Human STAT5B shares the identical sequence surrounding Tyr699.

Buffer

Purified IgG in 70 mM Tris (pH 8), 105 mM NaCl, 31 mM glycine, 0.07 mM EDTA, 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.

 

STAT5A/B phospho Tyr694/Tyr699 rabbit pAb tested by Western blot.
Nb2 nuclear extract (20 µg per lane) was probed with STAT5A/B phospho Tyr694/Tyr699 rabbit pAb (1:2,000).
    Lane 1: No treatment.
    Lane 2: Cells treated with prolactin (25 ng/ml for 20 minutes at 37°C).

STAT5A/B phospho Tyr694/Tyr699 rabbit pAb tested by dot blot analysis.
Dot blot analysis was used to confirm the specificity of STAT5A/B phospho Tyr694/Tyr699 rabbit pAb for STAT5A/B phospho Tyr694/699. Phosphorylated peptides corresponding to the immunogen and related peptides were spotted onto PVDF and probed with the antibody at 1:30,000. The amount of peptide (picomoles) spotted is indicated next to each row.

Background

STAT5A/B (signal transducer and activator of transcription 5) is a member of the STAT family of transcription factors. STATs undergo serine and tyrosine phosphorylation in response to growth factor or cytokine signaling. This phosphorylation results in dimerization and translocation of STAT proteins to the nucleus. The STAT5 protein is expressed in most cell types and is activated by multiple cytokines and growth factors including: IFNA1, IL-10, IL-6, IL-11, IL-12, IL-2, EGF, among others. Other members of the STAT family include STAT1α, STAT2, STAT3, STAT4, STAT5A, STAT5B and STAT6.

Positive Control

Active Motif's Nb2 nuclear extract (prolactin stimulated) (Catalog No. 36106) can be used as a positive control for STAT5A/B phospho Tyr694/Tyr699 antibody.
