Applications

Western Blot Validated

Application Notes

Applications Validated by Active Motif:
WB*: 1 - 2 µg/ml dilution

*Note: many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt / Sonication Protocol when preparing nuclear extracts for Western blot.

Immunogen

This EuHMT1 antibody was raised against a peptide derived from the N-terminal region of human EuHMT1.

Buffer

PBS pH 7.5 containing 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.

EuHMT1 antibody (pAb) tested by Western blot.
Nuclear extract of HeLa cells (20 µg) probed with EuHMT1 antibody (1 µg/ml dilution).

Background

EuHMT1 (Euchromatic Histone Methyltransferase 1, EHMT1, EuHMTase1, G9a like protein, GLP1) is a SET domain-containing histone methyltransferase specific for lysine 9 of histone H3. It is possible that this protein is involved in the silencing of Myc and E2F-responsive genes and thus could play a role in regulating cell cycle transitions from G0 to G1.

Positive Control

Active Motif's HeLa nuclear extract (Catalog No. 36010) can be used as a positive control for EuHMT1 antibody.

Storage

Some products may be shipped at room temperature. This will not affect their stability or performance. Avoid repeated freeze/thaw cycles by aliquoting items into single-use fractions for storage at -20°C for up to 2 years. Keep all reagents on ice when not in storage.
