Background
BRDT is similar to the RING3 protein family. It possesses two bromodomain motifs and a PEST sequence (a cluster of proline, glutamic acid, serine, and threonine residues), characteristic of proteins that undergo rapid intracellular degradation. The bromodomain functions as a ‘reader’ of epigenetic histone marks and regulates chromatin structure and gene expression by linking associated proteins to the recognized acetylated nucleosomal targets.

Protein Details
The peptide corresponding to amino acids 21-137 that contains the bromodomain sequences of BRDT (accession number NP_001717.2) was expressed in E. coli and contains an N-terminal His-Tag and C-terminal FLAG-Tag with an observed molecular weight of 20 kDa. It shows binding specificity for acetylated H2BK12/15, H3K9/14/18/23, H4K12, H4K5/8, H4K8/12, H4K12/16, H4K5/12, H4K5/8/12/16 and H4K5/8/12/16.

3.3 uM histone peptide H4K5/8/12/16(4Ac) was incubated with BRDT (21-137) protein in reaction buffer including 50mM HEPES-NaOH pH 7.0, 0.1% BSA overnight at room temperature. Anti-Flag antibody was used to detect reaction products.
