Immunogen
This STAT3 phospho Ser727 antibody was raised against a peptide containing phospho Ser727 of human STAT3.
Buffer
Purified IgG in 70 mM Tris (pH 8), 105 mM NaCl, 31 mM glycine, 0.07 mM EDTA, 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.
STAT3 phospho Ser727 pAb tested by Western blot.
Hep G2 nuclear extract (15 µg per lane) was probed with STAT3 phospho Ser727 pAb (1:1,000).
Lane 1: cells stimulated with IL-6 (100 ng/ml for 15 minutes at 37°C).
Lane 2: No treatment.
STAT3 phospho Ser727 pAb tested by dot blot analysis.
Dot blot analysis was used to confirm the specificity of STAT3 phospho Ser727 pAb for STAT2 phospho Ser727. Phosphorylated peptides corresponding to the immunogen and related peptides were spotted onto PVDF and probed with the antibody at 1:30,000. The amount of peptide (picomoles) spotted is indicated next to each row.

Background
STAT3 (signal transducer and activator of transcription 3) is a member of the STAT family of transcription factors. STATs undergo tyrosine and serine phosphorylation in response to growth factor or cytokine signaling. This phosphorylation results in dimerization and translocation of STAT proteins to the nucleus. The STAT3 protein is expressed in most cell types and is activated by multiple cytokines and growth factors including: IFNα, IL-10, IL-6, IL-11, IL-12, IL-2, EGF, among others.
Positive Control
Active Motif's Hep G2 nuclear extract (IL-6 stimulated, 100 ng/ml) (Catalog No. 36107) can be used as a positive control for STAT3 phospho Ser727 antibody.
Storage
