Application Notes
Applications Validated by Active Motif:
ChIP: 2.5 - 10 µg per ChIP
This antibody has been published, but has not been validated by Active Motif, for Western blot. For an antibody validated for use in Western blotting, see Catalog No. 39357.
This antibody is also available as an AbFlex® engineered recombinant antibody. For details on the corresponding AbFlex Recombinant Antibody, see Catalog No. 91163.
Published Applications
The following applications have been published using this antibody. Unless noted above, Active Motif may not have validated the antibody for use in these applications:

Immunogen
This Suz12 antibody was raised against a recombinant protein corresponding to full-length human Suz12.
Buffer
Purified IgG in 70 mM Tris (pH 8), 105 mM NaCl, 31 mM glycine, 0.07 mM EDTA, 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.
Suz12 antibody (mAb) tested by ChIP.
Chromatin IP performed using the ChIP-IT® Express Kit (Catalog No. 53008) and HeLa chromatin (1.5 x 106 cell equivalents per ChIP) using 10 µg of Suz12 antibody or the equivalent amount of rabbit IgG as a negative control. Real time, quantitative PCR (RT-qPCR) was performed on DNA purified from each of the ChIP reactions using a primer pair specific for the Myt1 gene promoter. Data are presented as Fold Enrichment of the ChIP antibody signal versus the negative control IgG using the ddCT method.
Background
Suz12 (Suppressor-of-Zeste 12 protein, also designated as ChET9, JJAZ1 protein) is a component of the Polycomb group complexes 2, 3 and 4 (PRC2, PRC3 and PRC4), which also contain the histone methyltransferase EZH2, EED (Embryonic Ectoderm Development protein) protein and the histone-binding proteins RbAP46 and RbAP48. PRC2 is responsible for di- and trimethylation of histone 3 at lysine 27 (Histone H3K27me2 and Histone H3K27me3), which leads to transcriptional repression of the target genes. Suz12 is a nuclear protein belonging to the VEFS family. Suz12 contains a zinc-finger binding motif and a VEFS box, which is required for the interaction between Suz12 and EZH2.
Positive Control
Active Motif's HeLa nuclear extract (Catalog No. 36010) can be used as a positive control for Suz12 antibody.
qPCR Primers
Active Motif's Human Positive Control Primer Set CCND2 (Cat. No. 71008) and Human Negative Control Primer Set 3 (Cat. No. 71023) have been validated for performing qPCR and endpoint PCR when this antibody has been used for ChIP on human samples. For ChIP experiments with this antibody on mouse samples, the Mouse Negative Control Primer Set 1 (Cat. No. 71011) has been validated for both qPCR and endpoint PCR.
Chromatin IP Assays
This antibody has been validated for use in ChIP and/or ChIP-Seq, and can be used with Active Motif's ChIP-IT® High Sensitivity Kit or our magnetic bead-based ChIP-IT® Express Kits.
